Yes. You can use your ticket on any day — not necessarily the calendar date you bought it on. The clock (75 min, 24 h, 48 h, 72 h, 7 days) only starts when you first validate it at the yellow Tappy reader.
Vaporetti keep a reduced "minimum service" on the main lines, and the night service runs as normal. Buses and trams have guaranteed slots from 06:00–08:59 and 16:30–19:29. There is no automatic refund — but a goodwill request through GetYourGuide is usually more flexible than going directly to ACTV.
Mostly no. Acqua alta is Venice's seasonal high-tide flooding, mainly between October and March, when the lagoon rises and parts of the city get wet feet. Vaporetti keep running through most of it — but specific stops close above certain tide levels: above 95 cm Line 3 is suspended, above 105 cm the Zattere stop towards San Marco and Lido closes, and above 120 cm access to some pontoons is blocked.
