From Piazzale Roma or Ferrovia, Line 3 reaches Murano Colonna in about 20 minutes. Direct, big boats. Catch only Monday to Friday, 06:21 to 19:18 — no service evenings or Sundays.
From Fondamente Nove (pontoon "A") to Murano Faro in about 10 minutes, every 20 minutes from 04:20 to 22:25. Continues on to Burano. The most flexible option.
From Fondamente Nove or any outer-ring stop, 4.1 and 4.2 swing past Murano on their full Venice loop. Slower but more scenic, all week, every 20 minutes.
Line 3 from Piazzale Roma: about 20 minutes. Line 12 from Fondamente Nove: 10 minutes to Murano Faro. Lines 4.1/4.2: about 20 minutes from Fondamente Nove, longer from elsewhere on the loop.
Yes — the Blue Line and Red Line both stop at Murano Colonna en route from Marco Polo Airport. But Alilaguna is NOT included in your Vaporetto ticket; it costs €10 single segment. Use the vaporetto if you are already in town.
